In this study, the lack of tetracycline-induced fluorescence upon 1-hour incubation in the presence of the triptorelin analogue was attributed to the formation of hydrophobic complex formation between tetracyclines and the triptorelin analogue and consequent inhibition of the tetracycline fluorescence.
The peptide analogues had a higher affinity for AT4 cells compared to that of tetracycline. The respective IC~50~ values were 28.3 ± 1.6 and 31.5 ± 1.9 *μ*mol/L for the analogues **14a** and **14b**, respectively, compared to 0.57 ± 0.05 *μ*mol/L for tetracycline. This may be attributed to the formation of a more compact, hydrophobic peptide–drug complex, which can lead to binding to proteins. This hypothesis is supported by the observed increase in the affinity of the analogues in comparison to the tetracyclines. The higher lipophilicity and the conformational rigidity of the cyclic peptides may also account for the improved binding affinity of the analogues compared to that of the linear peptides.
